Hsp70 chaperones consist of two functional domains: the 44 kDa Nucleotide Binding Domain (NBD), that binds and hydrolyses ATP, and the 26 kDa Substrate Binding Domain (SBD), which binds unfolded proteins and reactivates them, utilizing energy obtained from nucleotide hydrolysis. The structure of the SBD of the bacterial Hsp70, DnaK, consists of two sub-domains: A β-sandwich part containing the hydrophobic cavity to which the hepta-peptide NRLLLTG (NR) is bound, and a segment made of 5 α-helices, called the “lid” that caps the top of the β-sandwich domain. In the present study we used the Escherichia coli Hsp70, DnaK, as a model for Hsp70 proteins, focusing on its SBD domain, examining the changes in the lid conformation. We deliberately decoupled the NBD from the SBD, limiting the study to the structure of the SBD section, with an emphasis on the interaction between the charges of the peptide with the residues located in the lid. Molecular dynamics simulations of the complex revealed significant mobility within the lid structure; as the structure was released from the forces operating during the crystallization process, the two terminal helices established a contact with the positive charge at the tip of the peptide. This contact is manifested only in the presence of electrostatic attraction. The observed internal motions within the lid provide a molecular role for the function of this sub-domain during the reaction cycle of Hsp 70 chaperones.  相似文献

In this paper we summarize a study of the reactions of plasma-activated nitrogen with silane and NO admixture during the plasma-enhanced chemical vapour deposition (PECVD) of Si3N4 films outside the plasma region. The dominant role of nitrogen atoms, mainly in the ground (4S) state, was demonstrated experimentally. Probable chemical processes in the PECVD zone are discussed.  相似文献

We performed direct solvent-free amination of multi-walled carbon nanotubes (MWCNTs) with nonylamine, dodecylamine, octadecylamine, 4-phenylbutylamine and 1,8-ocanediamine at a temperature of 150-170 degrees C and reduced pressure. Thermogravimetric analysis and temperature-programmed desorption-mass spectrometry revealed that a major amine fraction decomposes in a temperature interval of 250-500 degrees C, thus existing on multi-walled carbon nanotubes as chemically bonded species; a minor amine fraction was found in physisorbed form. The new derivatization technique combines simplicity in implementation and attractive features of "green" chemistry. It requires no additional chemical activation, but thermal activation instead; it is relatively fast since it can be completed in about 2 h; the high temperature allows one to spontaneously remove excess amine from the nanotube and minimize the possibility of physical adsorption; there is no need to use an (organic) solvent medium. In the case of diamines (represented in this study by 1,8-ocanediamine), the functional groups introduced can be potentially used as chemical linkers for anchoring metal complexes and nanoparticles to multi-walled carbon nanotubes, for adsorption and concentration of trace metal ions.  相似文献

The “top-down” process via direct conversion of the micro (μm)-to-submicroscale (sub-μm) particle was applied in this work by using eutectic chloride salts to prepare BaZrO3. The particle size at optimum condition could be decreased by more than 10 times from 2.1 ± 0.9 μm to 168 ± 23 nm without destroying the 1:1 of Ba:Zr stoichiometry. The uniform sub-μm-BaZrO3 powder was sintered in order to obtain ~98% dense ceramic at 1400°C/10 h, which is significantly lower than the 1650°C in normal cases. The microwave dielectric constant, tan δ, and quality factor were also determined. Furthermore, this method also was applied to lead-free piezoelectric material in the 0.87BaTiO3–0.13BaZrO3–CaTiO3 (0.87BT–0.13BZ–CT) system. The particle size of 0.87BT–0.13BZ–CT was reduced greatly from >10 µm to 2.8 ± 0.4 µm. It can be proved that salt flux dissolution method enables high-purity with uniform sub-micro/nanometer powder production in one step by using simple laboratory equipment and low-cost raw materials.  相似文献

For Gadoid fishes, formaldehyde can be generated in tissues in huge amounts during endogenous enzymatic degradation of natural osmoprotectant trimethylamine-N-oxide. This paper describes two enzymatic methods for assay of formaldehyde in fish food products using alcohol oxidase (AOX) and formaldehyde dehydrogenase (FdDH) isolated from the thermotolerant methylotrophic yeast Hansenula polymorpha. AOX-based method exploits an ability of the enzyme to oxidise a hydrated form of formaldehyde to formic acid and hydrogen peroxide monitored in peroxidase-catalysed colorimetric reaction. In FdDH-based method, a monitored coloured formazane is formed from nitrotetrazolium salt during reduction by NADH, produced in formaldehyde-dependent reaction. It was demonstrated an applicability of both methods for assay of formaldehyde in fish products. The optimal protocols for analysis procedures have been elaborated and analytical parameters of both enzymatic methods have been established. The both methods were demonstrated that some fish products (hake and cod) contain high formaldehyde concentrations (up to 100 mg/kg wet weight).  相似文献
